Development and Validation of HPLC Method for Determination of Diclofenac Sodium by Tape Stripping Method

Abstract
Objective: To develop and validate the HPLC method for determination of Diclofenac sodium by tape stripping method.
Method: The chromatographic separation of Diclofenac was carried out using Waters Agilent chromatographic system, UV detector and X'terra RP18 (250*4.6) mm, 5µ column. After preparation of solutions, diluents (one injection), standard solution (five injections), and sample solution (duplicate injection) were injected into the chromatographic system. The calibration curves were obtained by plotting response versus concentration. The HPLC method was validated based on ICH guidelines with validation parameters viz. Retention time, Linearity and range, Accuracy, Precision.
Results: For the separation of Diclofenac by HPLC method ACN: potassium dihydrogen orthophosphate (70:30) pH 3.00 was selected as optimal mobile phase based on system suitability parameters with well resolved peaks of all formulations of Diclofenac. The wavelength for the detection of both compounds was 276 nm with best detector response. The linearity of the response of the drug was verified from 1 to 10 ìg/ml concentrations. The parameters like Retention time, Linearity and range, accuracy, precision were studied as validation parameters.
Conclusion: The proposed method can be used for determination of Diclofenac sodium in stratum corneum by tape stripping method.
