Anticholinergic property of ethanol extract of Moringaoleifera Leaves: An in vivo and in vitro approach

Solomon N. Ijioma, Chinenyenwa O. Nwosu, Charles Onyenegecha.

Abstract
Objective: The anticholinergic property of ethanol extract of Moringa oleifera leaf extract was investigated using in vivo and in vitro experimental models. Materials and Methods: For the in vivo work, 30 mice (20-25g) were divided into 6 groups of 5 mice each. Groups 2-6 were treated with Atropine (0.1mg/kg) and graded doses of Moringa oleifera leaf extract (50, 100, 200, 400mg/kg) respectively while group 1 served as control. Thirty minutes after treatment all animals were given 0.2ml oral dose of activated charcoal. All animals were sacrificed and dissected in a further 30 minutes to measure the distance moved by the charcoal meal against the full length of the small intestine. For the in vitro studies, 6 guinea pigs (320-400g) and 6 rabbits (1.8-2.5kg) were used. The effects of extract on acetylcholine induced contractions in isolated guinea pig ileum and rabbit jejunum were found. Results: All doses of the extract inhibited acetylcholine induced intestinal motility significantly (P
