Effect of green tea on ibuprofen-induced proximal tubular necrosis in kidney of adult rat

Abstract
Objective: To assess the effect of green tea on ibuprofen-induced proximal tubular necrosis in kidneys of adult rat.
Methodology: Thirty Sprague-Dawley rats, males and non-pregnant females, were selected and divided into three groups. Each group consisted of 10 animals. Group A served as control. Group B was given ibuprofen 120mg/kg bodyweight/day. Group C received green tea 1ml/100mg bodyweight/day in addition to ibuprofen 120 mg/kg. The dose was administered once a day for a period of 8 weeks. Twenty-four hours after administration of last dose, the animals were sacrificed and kidneys removed. Proximal tubular necrosis was assessed and graded in kidney specimens. The data were analyzed using SPSS version 22. Results: No proximal tubular necrosis was observed in control group A. In group B, medium necrosis was observed in one specimen (level II), severe necrosis in six specimens (level III) and extra severe necrosis in three specimens (level IV). In seven specimens from group C, no necrosis (level 0) while three showed mild necrosis (level I).
Conclusion: Ibuprofen induced necrosis in proximal renal tubules and administration of green tea significantly prevented necrosis in renal proximal tubules.
